Jag har identifierat tre typer av det enzym som används för att kopiera DNA som fungerar bättre i närvaro av andra substanser än den enzymvariant som används som standard inom kriminaltekniken i dag.
Vad är syftet med din forskning?
Inom kriminaltekniken utgår man ofta från prover som innehåller relativt lite DNA och som dessutom är nedsmutsade av andra substanser. Det kan till exempel vara ett prov från en cigarettfimp som innehåller både papper och ämnen från tobaken. För att kunna analysera DNAt i provet kopierar man upp det med hjälp av enzymet DNA-ploymeras. Problemet är att DNA-plomeraset störs om det finns andra molekyler än DNA i provet. I dag försöker man därför rena sina prov så mycket som möjligt vilket är en arbetsam process. Att rena upp provet innebär också att man förlorar genetiskt material på vägen. Därför ville jag hitta ett mer robust sätt att hantera provet.

Jag har screenat olika DNA-polymeras och hittat tre stycken som fungerar bättre än det enzym som används över hela världen i dag. Jag har också optimerat buffert, pH och jonsammansättning för dessa tre enzym och testat dem på 32 prov i Statens Kriminaltekniska Laboratorium, SKLs, arkiv. Med dagens metod gick det inte att få fram några användbara DNA-profiler ur proven men med min metod gick det att använda 28 av proven.
Vad hoppas du dina resultat ska användas till?
Framför allt vill jag att folk ska få upp ögonen för att de standardmetoder man använder kanske inte alltid är de bästa. Ja blev själv förvånad över att det blev så bra resultat som det blev med min metod, man tänker sig liksom att reaktionen är som den är och att den inte går att förbättra. Det ska bli intressant att se vad vi får för respons från andra kriminaltekniska labb i världen eftersom det här är ett lite nytt tankesätt.
